Bench sync 29mm smoothies question?

When bench syncing 29mm smoothbores I find two screws at the top of the slide. One is threaded with a locknut and the other is just a normal 8 or 10mm bolt. I know the adjuster will raise and lower the slide, however do I need to loosen the other bolt as well or leave it be?

Just open the top nut that is around the small adjuster screw. Then adjust, then re-tighten nut.

I like to bench sync using a piece of metal coat hanger. I adjust the slide up high, then slide the rod through the bore and tighten down the slide to the rod just gently. Then I turn the carb on its side and slowly loosen the adjuster until the rod falls out on its own, then tighten the adjuster nut.

Do the same for each and they will be dead bang on.
