Tuning question

So what is the procedure to tune my bike to the best result. I never synchronize my carbs. I just looked and judged the opening by eye. But now that I am doing an engine rebuild I would like to do it right. I am looking for the steps in the procedure to do it. I noticed I was running really rich by my plugs in some cylinders but fine in the others. I have a 20 pilot and 117.5 mains. Not sure what the needle jet is set to. This is all for a Z1B with a denco header exhaust and K&N pod filters. Everything else is stock.
